Title: Pharmacological Evaluation of Herbomineral Formulation in Isoproterenol - Induced Myocardial Infraction

Abstract: AIM AND OBJECTIVE: Myocardial Infarction, is an irreversible damage of myocardial tissue due to prolonged ischemia and hypoxia. In the past few decades there has been an increasing demand for the herbal drug treatment for various ailments, and many plant drugs from traditional medicine like Ayurveda are being explored globally. The aim of present investigation was to evaluate cardioprotective activity of the herbomineral formulation using animal model of isoproterenol induced myocardial infarction. MATERIAL AND METHOD Healthy Sprague Dawley rats weighing 250-300 gm were grouped: normal control, disease control, disease control group pretreated with the formulation (100 mg/kg, p.o) and disease control pre-treated with the standard (propranolol, 10 mg/kg, p.o). After pre-treatment of 4 weeks, myocardial necrosis was induced by subsequent injection of ISO (85 mg/kg/day, s.c.) into rats. Serum biochemical parameters like glucose, lipid profile, CK-MB, LDH, SGOT, SGPT were evaluated. Various haemodynamic parameters like blood pressure, ECG were measured. Weight of the heart was noted down to calculate the index of hypertrophy such as left ventricular weight to body weight, (LV/BW), heart weight to body weight (HW/BW). Animals were sacrificed and heart was excised for histopathology, antioxidant study (MDA, SOD, GSH, Catalase, nitrite) and collagen estimation. RESULTS Formulation pre-treated rats showed significant protection against ISO induced elevation in serum biochemical parameters such as serum chloesterol, triglyceride, LDL, VLDL and serum level of cardiac marker enzymes like CK-MB, LDH and transaminase level (SGOT, SGPT) as compared to ISO control group. There was no significant change observed in blood glucose and HDL level. Treatment showed significant improvement in haemodynamic parameters such as ST elevation, QT interval, hypertension and also produced significant decrease (p≤0.05) in index of hypertrophy. Significant protection was observed in formulation treated heart in histopathology sections. Result from our study also showed that formulation could efficiently protect against ISO intoxicated myocardial necrosis by reducing collagen levels and endogenous antioxidant enzyme levels significantly.CONCLUSION Our data suggests that formulation may be beneficial for the treatment of myocardial damage associated with myocardial necrosis. Therefore it could be concluded that the formulation is having a cardio protective effect.
