dc.description.abstract | Breast cancer is the most prevalent disease among women in the world, Current scenario of breast cancer management is chiefly revolving around the presence of the diagnostic and predictive markers. The tumor with no presence of these markers is known as Triple-negative breast cancer and there is no treatment of these TNBC patients. Any cancer cells spread to distant organs which are known as metastasis which is true for Breast cancer also. However, it has been shown in various studies that breast cancer metastasizes in chiefly in four distinct organs and they are Lung, Brain, Bone, and liver. Taking together it is difficult to manage TNBC tumors and along with that if it metastasizes than it will be more difficult to treat. Hence, we intended to unearth the Transcriptomic analysis of TNBC patients. In the present study, we intend to identify the key genes responsible for bone metastasis using bioinformatics APPROACH. Subsequently, validation of the key genes will be done by using TNBC breast cancer cell lines. We have used various approaches and used numerous software (PANTHER, Network analyst, R package). Moreover, various databases like e (NCBI, GEO2R, Array express). Using meta-analysis covering 2500 targeted genes was used for comparison of gene expression levels. Gene expression profiling of breast cancer helped in the understanding of the heterogeneity of breast cancer at the genomic level. A panel (123) genes were found to be significantly differentially expressed. Five of upregulated genes are 1, 2, 3, 4, 5 which have been associated with tumor progression, angiogenesis, and metastasis of breast cancer. This will help for the earlier diagnosis of breast and to find a potential biomarker for breast cancer metastasis. Along with the above study, our aim is the identification of responsible mRNA/miRNA Using breast cancer cell line MCF -7(ER+, PR+) and MDA-MB-231 (Triple Negative Breast Cancer Cell line) are used and the growth curve is performed. In the current study, we will identify responsible mRNA/miRNA using a breast cancer cell line by transcriptome data analysis of bone metastasis in breast cancer by using a bioinformatics approach. The identification of circulating mRNA/miRNA will provide a unique opportunity for earlier disease identification and monitoring of disease. Moreover, we are trying to identify key genes that are used as a biomarker. Gene expression profiling of breast cancer helped in the understanding of the heterogeneity of breast cancer at the genomic level.
