Abstract: Black mustard and Flaxseed were purchased from marked and germinated. Screening for amylase in black mustard was done by starch agar plate assay, starch – iodine tube assay and zymography. Presence of Lipase in flaxseed was confirmed after getting positive results on olive oil agar, tributyrin agar plates and zymography. Quantitative analysis of amylase was performed by DNSA method. Various types of chromatography was done ( Ion- Exchange chromatography with DEAE sephadex A-50 and Q sepharose, Gel filtration chromatography with sephadex G -75 and gel filtration high performance liquid chromatography) to purify the proteins.
